Gut-Lumen Uricase × ABCG2 Genotype Stratification + Flux Model (comp-019)¶
Not decision-usable. COMP-019's code did not use the luminal-urate concentration or UOX Km stored in its inputs and assumed 24 hours of saturated activity. Separately, COMP-019's unconditional flat-dose classification is not robust to COMP-044's tested substrate-occupancy and finite-window diagnostics. COMP-044 supplies no replacement ΔSUA, dose, genotype order, physiological regime, efficacy model, topology/chassis selection, production-sufficiency target, or safety conclusion. The retired model remains recoverable through Git for provenance only; do not use it for dose or efficacy decisions.
What survives¶
Phase A found no Q141K-stratified uricase clinical outcome in the sources searched for comp-019 as of 2026-05-08. This is a bounded search result, not proof of universal absence. Q141K therefore remains a prospective stratification variable rather than a computationally established response predictor.
Current decision¶
The Phase B outputs are not eligible for dose, efficacy, genotype-order, or engineering decisions. The gut-lumen uricase track remains open; substrate, oxygen, peroxide, access, survival, transit, and topology require direct measurement. Exact configurations must first be built or supplied and characterized; validation experiment 1.33 then supplies the configuration-level comparison, and §1.36 precedes animal escalation.
