Koji Multi-Payload Strain Hypothesis

1. Configuration boundary

This track asks whether one engineered Aspergillus oryzae strain can produce active UOX and lactoferrin without either cassette, the host, or the process degrading the other output. A combined strain is not the assumed endpoint. It is justified only if both single-cassette legs pass independently and co-expression preserves their measured performance.

Native kojic acid, ergothioneine, and digestive-enzyme outputs are covariates to measure. Their presence in a parental strain does not establish useful exposure, combination benefit, or retention after engineering. Failure narrows or kills this configuration; it does not hold up the koji track or Open Enzyme.

2. Component evidence

2.1 UOX leg

The UOX leg inherits the unresolved gut-lumen constraints: physiological substrate supply, topology, oxygen, peroxide, access, processing, persistence, reabsorption, and systemic compensation. No active-UOX titer, serving, human dose, or serum-urate effect has been established for an A. oryzae configuration.

Sequence and topology remain experimental variables. A. flavus Q00511, disclosed Candida utilis derivatives, and other candidates enter a matched screen; none is the default oral sequence. In Vitro and Mechanistic Extrapolation. See UOX variant selection and validation §1.33.

2.2 Lactoferrin leg

Ward et al. reported recombinant human lactoferrin from A. oryzae at 25 mg/L in submerged culture in 1992. In 1995, an A. awamori glucoamylase–KEX2 fusion plus strain improvement exceeded 2 g/L in a different submerged process. Sun et al. later reported a 2.2 Å structure of the A. awamori-produced protein consistent with native human lactoferrin. In Vitro. These are strong single-protein expression priors, not a solid-state-koji titer, a dual-cassette result, or an exposure target.

Lactoferrin has gout-adjacent mechanistic evidence: binding to lipid A and soluble CD14 was measured in vitro; NLRP3/caspase-1 and GSDMD-related effects were reported in non-gout animal and cell systems. In Vitro + Animal Model. These findings support testing a defined mechanism. They do not establish efficacy from a koji-derived product or additive benefit with UOX. The evidence home is lactoferrin.

2.3 Native kojic-acid output

Parental A. oryzae can produce kojic acid under some strain and process conditions. The experiment must measure it against parental and single-cassette controls rather than count it as an automatic payload. In Vitro; useful exposure and combined benefit unmeasured.

2.4 Native ergothioneine and digestive-enzyme outputs

Parental A. oryzae can produce ergothioneine and digestive enzymes under some strain and process conditions. Those outputs are substrate-, strain-, and process-dependent. The dual-cassette experiment must measure them against the parental and single-cassette controls rather than count them as automatic payloads. 2.5 Optional carnosine leg

Carnosine is outside the active dual-cassette design. Its androgen–urate rationale composes separate animal-model observations and has not been tested as a combined androgen × carnosine mechanism. Any A. oryzae build requires its own exact construct, precursor-supply, titer, process-retention, exposure, and safety evidence. Validation §1.24 owns the current test.

3. Staged engineering test

3.1 What the Ward precedent establishes

Ward 1992 and 1995 plus Sun 1999 establish that Aspergillus hosts can produce structurally characterized human lactoferrin under the reported single-protein submerged processes. They do not establish solid-state output, dual-cassette compatibility, or a useful exposure target.

3.3 Multi-cassette calibration boundary

Published Aspergillus work supports integration of multiple copies and loci, but most examples repeat one protein or optimize one secretion program. They do not establish simultaneous production of two different active proteins with distinct folding, processing, localization, and assay requirements. In Vitro precedent; dual-payload performance unmeasured.

3.4 Engineering question and staged experiment

The combined configuration can fail through mechanisms that a single-cassette result will not reveal:

  • competition for transcription, translation, ER folding, secretion, KEX2-family processing, redox capacity, or vesicular traffic;
  • altered UOX localization, oligomerization, substrate access, or peroxide exposure;
  • lactoferrin truncation, misprocessing, aggregation, altered iron loading, or loss of activity;
  • integration instability or unequal copy-number retention;
  • host stress, growth loss, or altered native-metabolite and enzyme output;
  • process-specific losses during solid-state growth, harvest, drying, storage, or simulated transit.

Bulk protein mass or transcript abundance cannot close these questions. The decision variables are active products in their intended fractions after the complete process.

Validation §1.9 owns the staged comparison.

Stage A — lactoferrin alone

Implement a provenance-bound human-lactoferrin construct and compare direct secretion with a Ward-style carrier/processing design only where the exact host and process make that comparison meaningful. Measure identity, intact mass, processing, soluble and active output, aggregation, host stress, and batch reproducibility.

This stage may run while the §1.5 UOX build and §1.33 screen are pending because it makes no UOX claim.

Stage B — UOX alone

Run only after §1.33 retains a koji-compatible topology. Implement that topology in the intended A. oryzae process and measure UOX identity, active oligomer, localization, substrate-to-product conversion, oxygen, H₂O₂, viability, processing retention, and batch reproducibility.

Expression yield alone does not pass Stage B and cannot be converted into a serving or dose.

Stage C — dual cassette

Build the combined strain only after Stages A and B pass. Compare:

  1. parental host;
  2. empty-vector or marker control where appropriate;
  3. lactoferrin-only strain;
  4. UOX-only strain;
  5. dual-cassette strain;
  6. at least two independently verified integrants per construct.

Measure both products with the same assays used in their single-cassette stages. Add integration identity and copy number, transcript as a diagnostic, host growth and stress, viability, secretory-pathway burden, native kojic acid and ergothioneine, digestive-enzyme activity, and process retention.

Set promotion margins only after an assay-precision pilot. A dual strain advances when both active outputs remain reproducible relative to their single-cassette controls without a new peroxide, viability, stability, native-output, or safety failure. A favorable average cannot hide failure of one leg.

3.5 Cordycepin and other additional cassettes

Cordycepin is not part of this active configuration. It and any other added cassette require an independent mechanism, construct, burden, exposure, and single-cassette gate before interaction testing.

4. Failure branches

4.1 Two-strain or separate-production fallback

If both single-cassette legs pass but coexistence fails, a two-strain or separately produced comparison may be tested only when co-delivery retains a defined advantage. These are fallback experiments, not automatically shippable intermediates; they inherit mixture compatibility, exposure, impurity, release, safety, and manufacturing questions.

UOX fails §1.33 or §1.36 Do not rescue the configuration with lactoferrin, native metabolites, or more expression. Redirect the UOX leg or retire it.
Lactoferrin lacks retained activity or relevant target engagement Remove it from the configuration even if its titer is high.

7. Safety, translation, and falsification

The parental organism's food-use history does not transfer automatically to the engineered strain, payloads, impurities, viable-organism format, or intended exposure.

Before animal escalation, the UOX leg must pass §1.36. The combined configuration also requires construct identity, containment, contamination, impurity, allergenicity/immunogenicity, barrier, host-response, and repeat-exposure evaluation appropriate to its actual format.

The result is an assay-qualified research configuration, not a product, serving, or dosing protocol.

Falsification boundary

The one-strain hypothesis fails if reproducible co-expression cannot preserve both active products and acceptable host/process behavior, or if either component fails its own physiological, safety, or target-engagement gate. Document the failure mechanism and remove the unsupported configuration from the portfolio.

Success would establish only that one defined dual-cassette strain works under the measured process. It would not establish human exposure, clinical benefit, superiority to separate delivery, or that koji is the project's endpoint.
